General description

JMJD2C, also known as Lysine-specific demethylase 4C, and encoded by the gene KDM4C/ GASC1/ JHDM3C/ JMJD2C/ KIAA0780, is a histone demethylase that specifically demethylates ′Lys-9′ and ′Lys-36′ residues of histone H3, thereby playing a central role in histone code. JMJD2C does not demethylate histone H3 ′Lys-4′, H3 ′Lys-27′, nor H4 ′Lys-20′. JMJD2C can also demethylate trimethylated H3 ′Lys-9′ and H3 ′Lys-36′ residues, but JMJD2C has no activity on mono- and dimethylated residues. JMJD2C is overexpressed in several esophageal squamouse cancer cells and serves as a cancer marker for such cancers. This gene is also amplified in squamous cell carcinoma 1 protein (GASC-1). By demethylating H3K9me, JMJD2C/KDM4C acts as a transcription promoter on many target genes including genes the affect cell growth in normal or cancer cells. JMJD2C also regulates sphere formation by mediating the cross talk between Wnt and Notch pathways in colonic cancer cells and thus is a target for anti-cancer therapy for this disease.
